I'll give two ideas but I'll leave room for other people to give their own ideas.
Idea 1- The Lesser Sex
In is the year 2780. A strange meteor struck the Earth centuries ago, transforming the planet and combining the future with the ancient magical past. However, the meteor caused a side-effect, the entire female human population underwent a bizarre transformation, becoming the monsters from mythology and folklore. The transformation also made them grow in size until the smallest giantess was 1500ft tall. The giantesses could still breed with the tiny human males, however, the offspring would only be human if it were male and only be a monster if it were female. Obviously this meant that women were in charge and all government positions were filled by the giantess monsters. While most of the time, the titanesses got along well with their tinier male counterparts, some titanesses aren't so friendly.
Idea 2- The New Nuke
In this scenario, several women were abducted and experimented on by a fiendish scientist who transformed them into mutants resembling the creatures of mythology; manticores, pegasi, cerberuses, everything. The mutates were locked up until an experimental bomb went off, turning them to giant size. Now that these monster giantesses were released into the world, they seek revenge for their harsh treatment.
