Uo met Tohru at middle school. Uo lived up to her stereotypee as a Yankee, being the member of a street gang, and as such saw Kyoko as a legendary figure to fear and respect. She envisioned her duahgter to be a formidable force of immense strength. When she first set eyes on Tohru she first thought that she was an exchange student or a visitor from another part of Japan, since Tohru seemed too cheery to her to be Kyoko's daughter.
Tohru's innocence was surprising and Uo was shaken by both Tohru's joviality and her size. After a few days of getting to know one another Tohru carried Uo back to her place where Uo set eyes on Kyoko. The conversation between idol and disciple was rather unorthodox as Uo could not understand why Kyoko was constructing buildings now instead of destroying criminals; however, when Kyoko heard that Uo belonged to a street gang (more importantly the exact one), once Tohru was out of the room, Kyoko offered to temporarily return to executing. In other words, she insinuated to Uo that she was to be giantess food. After that shocking experience Uo found that Kyoko has softened up a bit for her daughter and started to like this way of life. For her, the rest is history.
Hana, on the other hand, had transfered to Tohru's middle school in the last year after a disturbing experience in her old school. Hana did not show any fear of Tohru at all when they met and even talked with her about their experiences, since they had both experienced prejudice. It did not take long for them to become good friends and Tohru even offered herself as a sort-of bus service for her friends, letting them ride on her shoulders as she strode to school.
